تعیین برخی از خصوصیات بیولوژیکی و مولکولی جدایه ارومیه ویروس موزاییک هندوانه

چکیده

ویروس موزاییک هندوانه یکی از شایع ترین ویروس ها روی کدوییان می باشد. به منظور ردیابی و تعیین پراکنش این ویروس در مزارع منطقه ارومیه، 326 نمونه از گیاهان کدو، هندوانه، خربزه و خیار دارای علایم موزاییک و رگبرگ نواری و غیره از 22 روستای منطقه ارومیه جمع آوری و با آنتی بادی اختصاصی WMV در آزمون DAS-ELISA مورد بررسی قرارگرفتند. از این تعداد، 94 نمونه آلوده تشخیص داده شدند. جهت بررسی دامنه میزبانی از گیاهان متنوعی متعلق به چهار خانواده استفاده گردید که گیاهان Cucurbita pepo، Cucumis melo L.cv.Felexusus و Chenopodium amaranticolor Coste et Reyn، Cucumis sativus L.cv. Peto Seed و Nicotiana tobacum cv. Burly به عنوان میزبان‌های ویروس تشخیص داده شدند. یک قطعه 966 نوکلئوتیدی با آزمون IC-RT-PCR تکثیر و مستقیماً برای تعیین ترادف فرستاده شد. در درخت های فیلوژنتیکی ترسیم شده جدایه ارومیه داخل هیچ‌کدام از 6 گروهی که قبلاً توسط محققین دیگر تعیین شده بود قرار نگرفت.

چکیده [English]

Watermelon mosaic virus is the most prevalent potyvirus infecting cucurbits in Iran. In order to determine the distribution and frequency of the virus, 326 watermelon, melon, cucumber, winter and summer squash samples were collected from 22 villages around Urmia during summer 2010 and tested by DAS-ELISA with a polyclonal antibody. Ninety four samples were detected as infected by ELISA. To determine host range of the virus, different plants from 4 familieas were tested. Cucurbita pepo, Cucumis melo L. Cv. Flexus, C.sativus L.cv. Peto Seed and Nicotiana tobaccum cv. Burley were detected as the virus hosts. One strain of squash that was positive in ELISA was used for IC-RT-PCR. A 958 bp product was amplified by IC-RT-PCR using specific primers and was sent for sequencing directly. Phylogenetic trees were constructed by Maximum likelihood, Maximum parsimony and Neighbour joining. Urmian isolate of WMV was not classified in any of the 6 groups which were shown by previous researches.
