مطالعه بیان ژن‌های SUT1، Chi3 و CYP1 در گوجه‌فرنگی پارازیت شده با گل جالیز (Orobanche aegyptica)

چکیده

گل جالیز (Orobanche spp) گیاه انگلی مطلق بوده که به ریشه بسیاری از گیاهان زراعی مهم همچون گوجه فرنگی، آفتابگردان، خیار، توتون و متصل شده و باعث کاهش قابل توجهی در عملکرد گیاه زراعی به ویژه در مناطق گرم و خشک همچون اروپا، آفریقا و آسیا می‌شود. تاکنون روش‌های متعددی برای کنترل گل جالیز استفاده شده است، اما عموماً این روش‌ها گران بوده و موثر عمل نمی‌کنند. درک بهتر نحوه واکنش گیاه میزبان و همچنین مکانیسم مولکولی مقاومت به این گیاه انگل به منظور یافتن روش‌های بهتر جهت کنترل آن ضروری به نظر می‌رسد. در این مطالعه بررسی بیان دو ژن کدکننده آنزیم های کیتیناز (Chi3) و سیستئین پروتئیناز (CYP1)، و همچنین یک ژن کدکننده انتقال دهنده ساکارز (SUT1)، در واکنش گیاه گوجه فرنگی به آلودگی با گل جالیز، در دو ژنوتیپ متحمل و حساس با استفاده از تکنیک Real time PCR انجام شد. هر سه ژن در ساعات اولیه پس از آلودگی افزایش بیان را نشان دادند. بیان انتقال دهنده ساکارز و همچنین آنزیم سیستئین پروتئیناز در هنگام تنش در دو ژنوتیپ با یکدیگر تفاوت معنی‌دار داشته اما در مورد آنزیم کیتیناز این تفاوت مشاهده نشد. نحوة بیان ژن ها در دو ژنوتیپ حساس و مقاوم نشان داد که فعالیت های دفاعی گیاه گوجه فرنگی از همان ساعات اولیه پس از آلودگی شروع شده و رقم متحمل پاسخ دفاعی سریع تری به علائم منتقل شده از گیاه انگل می دهد. همچنین احتمالاً آنزیم های تجزیه کننده پروتئین (پروتئینازها)، یکی از ابزارهای دفاعی زودهنگام گیاهان در جلوگیری از نفوذ گل جالیز به ریشه می باشد.

چکیده [English]

Broomrape (Orobanche aegyptica) is an obligate root parasite causing yield losses in large number of crops such as tomato, sunflower, cucumber, tobacco. In tropical and Mediterranean countries from Europe, Africa and Asia. Several methods have been suggested for control of this parasite, but most of them are expensive and ineffective. For the efficient control, a better understanding of its interaction with host plant characterization of resistance mechanisms at molecular level is required, The expression of genes encoding chitinase, cysteine proteinase and succrose transporter in response to Orobanche aegyptica was studied in a tolerant and sensetive tomatos cultivars by Realtime PCR method. Results showed that the genes were up-regulated in early stage of infection. Two species also showed different expression in succrose transporter and cysteine proteinase, but the same expression was observed for chitinase gene. The behaviour pattern of two genotypes showed that defence activity of tomato started at early hours of infection and the resistant genotype responses were earlier than sensetive genotype to signal of invasion. Results also indicated that protease enzymes can be one of the important tools of plants for preventing the penetration and connection of parasite to its root.
