ردیابی ویروس S سیب زمینی (PVS) با استفاده از روش های سرولوژیکی و آزمون اختصاصی RT-PCR در استان های خراسان رضوی و همدان

چکیده
ویروس S سیب زمینی متعلق به جنس Carlavirus و خانواده Flexiviridae می باشد که دارای ذرات رشته ای خمیده به ابعاد 12 ×650 نانومتر و ژنوم RNA تک رشته ای مثبت می باشد. این ویروس از متداول ترین ویروس هایی است که تمام واریته های سیب زمینی موجود در دنیا را آلوده می کند. گونه های حساس به PVS در سه خانواده Solanaceae ،Chenopodiaceae و Amaranthaceae یافت می شوند. این ویروس از طریق مایه زنی مکانیکی و همچنین توسط شته سبز هلو Myzus persicae به صورت ناپایا منتقل می شود. به منظور ردیابی ویروس در منطقه، از حدود 50 مزرعه استان های خراسان رضوی و همدان، تعداد 555 نمونه از غده های گیاهانی که علایمی نظیر موزاییک، لکه-های نکروتیک، فرورفتگی جزئی رگبرگ، رگوز، تموج حاشیه برگ، پژمردگی و کوتولگی را نشان می دادند، جمع آوری شدند. سپس غده ها جهت گذراندن دوره خواب در دمای C◦4 قرار گرفتند. پس از جوانه زنی، با استفاده از آنتی سرم پلی کلونال، اختصاصی PVS در آزمون سرولوژیکی DAS-ELISA، غده های آلوده به این ویروس شناسایی شدند. عصاره تعدادی از نمونه هایی که در آزمون الایزا مثبت ارزیابی شدند، بر روی گیاهان آزمون شامل Chenopodium quinoa و amaranticolor Chenopodium ( لکه های کلروتیک روی برگ)، Nicotiana debneyi (موزاییک)، N. tabacum var. xanthi ( مصون)، Lycopersicon esculentum ( موزاییک) مایه زنی شد. نتایج آزمون الایزا نشان داد که تعداد 69 نمونه به این ویروس آلوده بوده اند. به منظور تأیید نتایج الایزا در نمونه های آلوده از روشRT-PCR و آغازگرهای اختصاصی جهت تکثیر ژن کد کننده پروتئین پوششی ویروس استفاده شد. آغازگرهای اختصاصی PVS-F و PVS-R، قطعه ای به طول 1118 جفت باز مربوط به ژن پروتئین پوششی ویروس را تکثیر نمودند.

Abstract
Potato virus S is a member of the genus Carlavirus in the family of Flexiviridae, with curved filamentous particles 650×12 nm and positive single stranded RNA genome. This virus is one of the most common viruses that infected all of potato varieties in the world. Susceptible host species belong mainly to the families Solanaceae, Chenopodiaceae and Amaranthaceae. Potato virus S transmits by mechanical inoculation and Myzus persicae as well in a non-persistent manner. Samples of 555 tubers showing symptoms of mosaic, necrotic spots, vein partial deepening, rugosity, undulation of the margin, wilting and dwarfing from 50 fields in Khorasan Razavi and Hamedan provinces were collected. After tubers passed dormancy period at 4˚C and germinated, PVS polyclonal and special antiserum were used in DAS-ELISA to identify infected potato tubers. Sap of infected samples inoculated on indicator plants such as Chenopodium quinoa and C. amaranticolor (chlorotic spots on the leaf), Nicotiana debneyi (severe mosaic), N. tabacum var. xanthi (immune) and Lycopersicon esculentum (mosaic). Results of ELISA test indicated that 69 samples out of 555 samples were infected. To confirm the results of ELISA test, RT-PCR and specific primer for amplification of coat protein gene were used. Specific primers amplified 1118 bp fragment related to coat protein of the virus.
