بررسی مولکولی شش جدایه ویروس زردی چغندر قند (Beet yellows virus) از استان‌های آذربایجان غربی، کرمانشاه و همدان بر اساس ناحیه پروتئین پوششی

چکیده

ویروس زردی چغندرقند (Beet yellows virus, BYV) متعلق به جنس Closterovirus و خانواده Closteroviridae از مهم‌ترین ویروس‌های چغندرقند به شمار می‌آید که در سراسر دنیا گسترده است. به منظور تعیین برخی از خصوصیات مولکولی این ویروس، در سال 1391 تعداد 75 نمونه برگی دارای علائم از مزراع عمده این محصول در استان‌های آذربایجان غربی، کرمانشاه و همدان جمع‌آوری شد. نمونه‌ها دارای علائمی نظیر زردی، رگبرگ روشنی و ضخیم و شکننده شدن برگ، بودند. نمونه‌های جمع آوری شده توسط آزمون سرولوژیکی الایزا و آزمون مولکولی RT-PCR مورد بررسی قرار گرفتند. در آزمون الایزا با استفاده از آنتی‌بادی اختصاصی آلودگی در 6 نمونه تایید شد. قطعه‌ای مربوط به پوشش پروتینی به طول bp615 در آزمون مولکولی با استفاده از آغازگرهای اختصاصی طراحی شده BYV-F/R، تکثیر و در ژل آگارز 1 درصد الکتروفورز گردید. نتایج تعیین توالی و مقایسه توالی‌های بدست آمده با جدایه‌های موجود در NCBI به وسیله نرم افزار DNAMAN7 نشان داد که جدایه‌های تعیین توالی شده دارای 99-89 درصد شباهت با جدایه‌های موجود در بانک ژن هستند. بیش‌ترین درصد شباهت جدایه‌های ایرانی در سطح نوکلئوتیدی (19/99 درصد) و آمینو اسیدی (55/97 درصد)، با جدایه ای از اوکراین (X73476) می‌باشد. آنالیز فیلوژنتیکی جدایه‌ها به وسیله نرم افزار MEGA5 انجام شد که طی آن جدایه‌های ایرانی در کنار جدایه اوکراین در یک گروه قرار گرفتند.

Beet yellows virus (BYV) belongs to the genus Closterovirus, Closteroviridae family, is one the most important viruses of sugar beet worldwide. In order to identify and characterize some molecular aspects of the BYV, a total of 75 symptomatic leaf samples of sugar beet were collected from sugar beet field of West Azarbayjan, Hamedan and Kermanshah provinces, Iran. Leaves showing general yellowing, vein clearing and thickness were collected. The primary detection of BYV performed by DAS-ELISA, using specific antibodies raised against coat protein of the virus, resulted in positive reaction of six out of 75 samples. The coat protein ORF of BYV isolates was amplified by RT-PCR, using BYV-F/BYV-R primer pair. Amplified fragments in expected size of 615 bp were directly sequenced. All sequence data were multiple aligned with other BYV isolates (from NCBI) using DNAMAN 7 software. An Ukrainian isolate (X73476) showed the highest nucleotide (99.19%) and amino acid (97.55%) identity with our isolates in this study. Phylogenetic analyses were conducted by MEGA 5.2. The results of phylogenetic analysis showed that BYV Iranian isolates and BYV Ukrainian isolate are clustered together.
