The virus is the second most important virus of Carnation around the world and the only DNA virus among infecting viruses of carnation. The virus infected plants were collected from greenhouse in Chenaran and two fragments of ~1500 bp and ~1000 bp respectively for coat protein and movement protein were amplified by PCR using specific primers for coat protein and movement protein genes of CERV. The iranian isolate was compared with two isolates of CERV (India and Holand) and some of caulimoviruses. After multiple sequence alignment the phylogenetic tree of coat protein (CP) and movement protein (MP) sequences of the CERV isolates were drawn by MEGA5 and DNAMAN softwares using Neighbor joining method. The results showed that the CP of the iranian isolate ( Acc. No. JF957838) has 98.9 and 99.3 percent identity with Dutch isolate (Acc. No. XO4658) in nucleotide (nt) and amino acid (aa) sequences respectively.While the CP of the iranian isolate has 98.3 and 98.6 percent identity in nt and aa sequences respectively. Also the MP of the iranian isolate (Acc. No. JF957839) has 97.7 and 99.6 percent identity with Dutch isolate (Acc. No. XO4658) in nt and aa sequences respectively. The MP gene of the iranian isolate has 96.1 and 98.9 percent identity with Indian isolate (Acc. No. AJ853858) in nt and aa sequences respectively. This is the first molecular study of CERV in Iran}, keywords = {Carnation etched ring virus,Phylogenetic analysis,Dianthus caryophyllus}, title_fa = {مقایسه فیلوژنتیکی جدایه ایرانی ویروس خراشک حلقوی میخک با جدایه‌های دنیا بر اساس ترادف ژن پروتئین پوششی و ژن پروتئین حرکتی}, abstract_fa = {ویروس خراشک حلقوی میخک (Carnation etched ring virus, CERV)، عضو جنس Caulimovirus و خانوادة Caulimoviridae است. این ویروس به عنوان دومین ویروس میخک از نظر پراکنش در دنیا و تنها ویروس DNA دار گزارش شده از آن می‌باشد. بوته‌های آلوده به این ویروس از گلخانه چناران جمع‌آوری و با استفاده از جفت آغازگرهای اختصاصی طراحی شده برای نواحی حفاظت شده از ژن پروتئین ‌پوششی و ژن پروتئین‌حرکتی ویروس، به ترتیب قطعاتی در اندازه تقریبی1500 و 1000جفت ‌باز تکثیر گردید. این جدایه به همراه دو جدایه موجود از این ویروس (هند و هلند) و نیز تعدادی از دیگر ترادف‌های کائولیموویروس‌ها در بانک ژن مقایسه و پس از همردیف‌سازی چند‌گانه درخت فیلوژنتیکی آن‌ها برای ژن پروتئین‌ پوششی و ژن پروتئین‌حرکتی CERV با روش Neighbor joining و با استفاده از نرم افزار DNAMAN و MEGA5 ترسیم گردید. این آنالیز فیلوژنتیکی نشان داد که در مورد ژن پروتئین‌پوششی شباهت جدایه ایرانی در سطح نوکلئوتیدی و آمینواسیدی به ترتیب با جدایه هلند 9/98 و 3/99 درصد و با جدایه هند به ترتیب 3/98 و 6/98 درصد می‌باشد. هم‌چنین در مورد ژن پروتئین‌حرکتی شباهت جدایه ایرانی در سطح نوکلئوتیدی و آمینواسیدی با جدایه هلند به ترتیب 7/97 و 6/99 درصد و با جدایه هند به ترتیب 1/96 و 9/98 درصد نشان داده ‌شد. این اولین گزارش از مطالعات مولکولی درباره ویروس خراشک حلقوی میخک در ایران است.}, keywords_fa = {ویروس خراشک حلقوی میخک,آنالیز فیلوژنتیکی,Dianthus caryophyllus}, url = {https://jpp.um.ac.ir/article_35249.html}, eprint = {https://jpp.um.ac.ir/article_35249_3228d109278c783326cb067060c14078.pdf} }
