For biolobical control of this disease,130 strains of Fluorescent Pseudomonads were isolated from rhizospheric soil of wheat in different regions of Khorasan province. Selection the more active strains was released, using dual culture test by Fluorescent Pseudomonads and Ggt on KB and PDA medium. Among them, 21 strains were selected as more active on Ggt showing the inhibited of mycelial growth at the rate of 11.25 up 51.25 %. The PCR technique was completed by using primers PCA2a and PCA3b for detection the gene responsible of Phenazine-1-carboxylic acid production. The result showed that 12 strains of 21 strains contain the Phenazine-1-carboxylic acid gene. The dark green pigmentation or crystalline deposits method was used for testing the expression of phenazine-1-carboxylic acid antibiotic. The results showed that 6 from 12 strains able to produce Phenazine-1-carboxylic acid. In greenhouse experiment the Phenazine-1-carboxylic acid positive strains showed higher ability of biocontrol on Take-all disease than other strains. This strains decreased disease severity at the rate of 77-80%. The comparable results were observed for fresh weight of root and aerial parts of plants. These results showed that Phenazine-1-carboxylic acid which produced by these strains has important role in reducing Take-all disease probably}, keywords = {Keywords: Biological control,Fluorescent Pseudomonads,Gaeumannomyces graminis var.tritici,Phenazine-1-carboxylic acid antibiotic,Take-all}, title_fa = {ردیابی آنتی‌بیوتیک فنازین- 1-کربوکسیلیک اسید در سودوموناس‌های فلورسنت جداسازی شده از ریزوسفر گندم و تاثیرآن در کنترل بیولوژیکی بیماری پاخوره‌گندم}, abstract_fa = {بیماری پاخوره‌گندم با عامل قارچی Gaeumannomyces graminis var. tritici (Ggt) یکی از مهم‌ترین بیماری‌های گندم در ایران به شمار می رود. به منظور کنترل بیولوژیکی این بیماری،130 جدایه سودوموناس فلورسنت از ریزوسفر گندم در نواحی مختلف استان خراسان جداسازی شد. برای انتخاب جدایه‌های سودوموناس فلورسنت با توانایی بازدارندگی از رشد قارچ، آزمون کشت متقابل باکتری‌ها با قارچ Ggt در محیط کشت سیب‌زمینی دکستروز‌آگار و کینگ ب انجام شد. از این میان، 21 جدایه با قابلیت بازدارندگی بین 25/51-25/11 درصد به عنوان جدایه‌های برتر از نظر ممانعت از رشد قارچ Ggt انتخاب شدند. برای ردیابی ژن بیوسنتز آنتی‌بیوتیک فنازین-1-کربوکسیلیک‌اسید ‌(PCA)‌، روش PCR با استفاده از پرایمرهای PCA2a و PCA3b انجام شد. نتایج نشان داد 12 جدایه از 21 جدایه انتخاب‌شده حاوی ژن بیوسنتز آنتی‌بیوتیک PCA بودند. جهت بررسی تولید این آنتی‌بیوتیک از روش کیفی تولید رسوب سبز تیره یا کریستالی در محیط کشت استفاده شد. نتایج نشان‌دهنده توانایی تولید این آنتی‌بیوتیک در 6 جدایه از 12 جدایه حاوی ژن بیوسنتز آنتی‌بیوتیک بود. در آزمایش‌های گلخانه‌ای نیز جدایه‌های تولید‌کننده آنتی‌بیوتیک فنازین-1-کربوکسیلیک‌اسید، نسبت به سایرین از توانایی بیشتری در کنترل بیماری پاخوره‌گندم برخوردار بودند و شدت بیماری را 80-77 درصد کاهش دادند. با کاربرد این جدایه‌ها همچنین افزایش قابل ملاحظه‌ای در وزن‌تر ریشه و بخش‌های هوایی گیاهان مشاهده شد. نتایج این تحقیق پیشنهاد می‌کند که احتمالا آنتی‌بیوتیک فنازین-1-کربوکسیلیک‌اسید تولید‌شده توسط جدایه‌های سودوموناس فلورسنت، در کاهش بیماری پاخوره در گندم نقش دارد.}, keywords_fa = {Keywords: Biological control,Fluorescent Pseudomonads,Gaeumannomyces graminis var.tritici,Phenazine-1-carboxylic acid antibiotic,Take-all}, url = {https://jpp.um.ac.ir/article_32229.html}, eprint = {https://jpp.um.ac.ir/article_32229_6bb605c13b0cc55364f1b783af6b2577.pdf} }
